Uncertain significance for DYRK1A-related intellectual disability syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001347721.2(DYRK1A):c.1970C>T (p.Ser657Leu), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces serine with leucine at codon 666 of the DYRK1A protein (p.Ser666Leu). The serine residue is highly conserved and there is a large physicochemical difference between serine and leucine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with DYRK1A-related conditions.
