Uncertain significance for Purine-nucleoside phosphorylase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000270.4(PNP):c.49C>T (p.Leu17Phe), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces leucine with phenylalanine at codon 17 of the PNP protein (p.Leu17Phe). The leucine residue is moderately conserved and there is a small physicochemical difference between leucine and phenylalanine. This variant has not been reported in the literature in individuals with PNP-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
