NM_025132.4(WDR19):c.106T>G (p.Tyr36Asp) was classified as Uncertain significance for Asphyxiating thoracic dystrophy 5; Senior-Loken syndrome 8 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs781578023, ExAC 0.01%). This sequence change replaces tyrosine with aspartic acid at codon 36 of the WDR19 protein (p.Tyr36Asp). The tyrosine residue is weakly conserved and there is a large physicochemical difference between tyrosine and aspartic acid. This variant has not been reported in the literature in individuals with WDR19-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
