NM_001768.7(CD8A):c.286_287delinsTT (p.Asp96Phe) was classified as Uncertain significance for Susceptibility to respiratory infections associated with CD8alpha chain mutation by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with CD8A-related conditions. This sequence change replaces aspartic acid with phenylalanine at codon 96 of the CD8A protein (p.Asp96Phe). The aspartic acid residue is moderately conserved and there is a large physicochemical difference between aspartic acid and phenylalanine. This variant is not present in population databases (ExAC no frequency).
