Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.168C>G (p.Asp56Glu), citing Ambry Variant Classification Scheme 2023: The p.D56E variant (also known as c.168C>G), located in coding exon 1 of the DNAH11 gene, results from a C to G substitution at nucleotide position 168. The aspartic acid at codon 56 is replaced by glutamic acid, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6000 samples (12000 alleles) with coverage at this position. This amino acid position is highly conserved on limited sequence alignment. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
