This variant lies in the RANBP2 gene (transcript NM_006267.5) at coding-DNA position 3028, where A is replaced by C; at the protein level this means replaces methionine at residue 1010 with leucine — a missense variant. Submitter rationale: This sequence change replaces methionine with leucine at codon 1010 of the RANBP2 protein (p.Met1010Leu). The methionine residue is highly conserved and there is a small physicochemical difference between methionine and leucine. This variant is present in population databases (rs758609757, ExAC 0.006%). This variant has not been reported in the literature in individuals with RANBP2-related conditions.
