This variant lies in the CSPP1 gene (transcript NM_001382391.1) at coding-DNA position 1645, where C is replaced by G; at the protein level this means replaces proline at residue 549 with alanine — a missense variant. Submitter rationale: This sequence change replaces proline with alanine at codon 544 of the CSPP1 protein (p.Pro544Ala). The proline residue is highly conserved and there is a small physicochemical difference between proline and alanine. This variant is present in population databases (rs754153035, ExAC 0.002%). This variant has not been reported in the literature in individuals affected with CSPP1-related conditions.
