Uncertain significance for Congenital sensory neuropathy with selective loss of small myelinated fibers — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002506.3(NGF):c.145C>A (p.Arg49Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with serine, which is neutral and polar, at codon 49 of the NGF protein (p.Arg49Ser). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with NGF-related conditions. ClinVar contains an entry for this variant (Variation ID: 962933). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
