Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.610C>A (p.Leu204Ile), citing Ambry Variant Classification Scheme 2023: The p.L204I variant (also known as c.610C>A), located in coding exon 5 of the RECQL gene, results from a C to A substitution at nucleotide position 610. The leucine at codon 204 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
