NM_024642.5(GALNT12):c.776T>G (p.Val259Gly) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V259G variant (also known as c.776T>G), located in coding exon 4 of the GALNT12 gene, results from a T to G substitution at nucleotide position 776. The valine at codon 259 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
