Uncertain significance for Charcot-Marie-Tooth disease type 4A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018972.4(GDAP1):c.1063C>T (p.Pro355Ser), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with GDAP1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces proline with serine at codon 355 of the GDAP1 protein (p.Pro355Ser). The proline residue is moderately conserved and there is a moderate physicochemical difference between proline and serine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0").
