NM_005228.5(EGFR):c.1024A>C (p.Ile342Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I342L variant (also known as c.1024A>C), located in coding exon 9 of the EGFR gene, results from an A to C substitution at nucleotide position 1024. The isoleucine at codon 342 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
