NM_000368.5(TSC1):c.2112T>G (p.Tyr704Ter) was classified as Pathogenic for Tuberous sclerosis 1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in TSC1 are known to be pathogenic (PMID: 10227394, 17304050). A different variant (c.2111_2112del) giving rise to the same protein effect observed here (p.Tyr704*) has been determined to be pathogenic (PMID: 9242607, 15595939, 26540169). This suggests that this variant is also likely to be causative of disease. This variant has not been reported in the literature in individuals with TSC1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Tyr704*) in the TSC1 gene. It is expected to result in an absent or disrupted protein product.
