NM_007194.4(CHEK2):c.340T>C (p.Trp114Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W114R variant (also known as c.340T>C), located in coding exon 2 of the CHEK2 gene, results from a T to C substitution at nucleotide position 340. The tryptophan at codon 114 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
