Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_017841.4(SDHAF2):c.337A>G (p.Ser113Gly), citing Ambry Variant Classification Scheme 2023: The c.337A>G variant (also known as p.S113G), located in coding exon 3 of the SDHAF2 gene, results from an A to G substitution at nucleotide position 337. The serine at codon 113 is replaced by glycine, an amino acid with similar properties. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will result in the creation or strengthening of a novel splice donor site. RNA studies have demonstrated that this alteration results in a transcript predicted to lead to a protein with an in-frame deletion of 11 amino acids; however, the exact functional impact of the deleted amino acids is unknown at this time (Ambry internal data). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
