Pathogenic for Leber congenital amaurosis 1; Cone-rod dystrophy 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000180.4(GUCY2D):c.65G>A (p.Trp22Ter), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in GUCY2D are known to be pathogenic (PMID: 10951519, 11328726). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. This variant has not been reported in the literature in individuals with GUCY2D-related conditions. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate insufficient coverage at this position in the ExAC database. This sequence change creates a premature translational stop signal (p.Trp22*) in the GUCY2D gene. It is expected to result in an absent or disrupted protein product.
