Uncertain significance for Congenital myasthenic syndrome 10; Fetal akinesia deformation sequence 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_173660.5(DOK7):c.241G>C (p.Val81Leu), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DOK7 gene (transcript NM_173660.5) at coding-DNA position 241, where G is replaced by C; at the protein level this means replaces valine at residue 81 with leucine — a missense variant. Submitter rationale: This sequence change replaces valine with leucine at codon 81 of the DOK7 protein (p.Val81Leu). The valine residue is weakly conserved and there is a small physicochemical difference between valine and leucine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with DOK7-related conditions. This variant is not present in population databases (ExAC no frequency).
