This variant lies in the NEFL gene (transcript NM_006158.5) at coding-DNA position 214, where G is replaced by T; at the protein level this means replaces aspartic acid at residue 72 with tyrosine — a missense variant. Submitter rationale: This sequence change replaces aspartate with tyrosine at codon 72 of the NEFL protein (p.Asp72Tyr). The aspartate residue is highly conserved and there is a large physicochemical difference between aspartate and tyrosine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with NEFL-related conditions.
