This variant has been observed in individual(s) with clinical features of PURA-related conditions (PMID: Invitae). In at least one individual the variant was observed to be de novo. For these reasons, this variant has been classified as Pathogenic. This sequence change results in a premature translational stop signal in the PURA gene (p.Tyr240*). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 83 amino acids of the PURA protein.
