Pathogenic for Arrhythmogenic cardiomyopathy with wooly hair and keratoderma; Arrhythmogenic right ventricular dysplasia 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004415.4(DSP):c.5460_5472del (p.Val1821fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 5460 through coding-DNA position 5472, deleting 13 bases; at the protein level this means shifts the reading frame starting at valine residue 1821, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change results in a premature translational stop signal in the DSP gene (p.Val1821Thrfs*12). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 1051 amino acids of the DSP protein. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with DSP-related conditions. This variant disrupts the C-terminus of the DSP protein. Other variant(s) that disrupt this region (p.Thr2634Lysfs*4, p.Arg2166*) have been determined to be pathogenic (PMID: 11063735, 27532257, 23671136). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. For these reasons, this variant has been classified as Pathogenic.
