Pathogenic for Osteogenesis imperfecta type I — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.11:g.50192025del, citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This variant has been observed in an individual affected with osteogenesis imperfecta type I (PMID: 30614853). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. Loss-of-function variants in COL1A1 are known to be pathogenic (PMID: 7942841, 9295084, 9443882). For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Gly662Valfs*104) in the COL1A1 gene. It is expected to result in an absent or disrupted protein product.
