This variant lies in the KAT6B gene (transcript NM_012330.4) at coding-DNA position 4538, duplicating one base; at the protein level this means shifts the reading frame starting at lysine residue 1514, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change results in a premature translational stop signal in the KAT6B gene (p.Lys1514Glufs*27). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 560 amino acids of the KAT6B protein. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with KAT6B-related conditions. This variant disrupts the C-terminus of the KAT6B protein. Other variant(s) that disrupt this region (p.Arg1797*) have been determined to be pathogenic (PMID: 22077973).
