Pathogenic for McKusick-Kaufman syndrome; Bardet-Biedl syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_170784.3(MKKS):c.47del (p.Pro16fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in MKKS are known to be pathogenic (PMID: 12107442, 20177705). This variant has not been reported in the literature in individuals with MKKS-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Pro16Hisfs*2) in the MKKS gene. It is expected to result in an absent or disrupted protein product.
