Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000368.5(TSC1):c.856C>G (p.Pro286Ala), citing Ambry Variant Classification Scheme 2023: The p.P286A variant (also known as c.856C>G), located in coding exon 7 of the TSC1 gene, results from a C to G substitution at nucleotide position 856. The proline at codon 286 is replaced by alanine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
