Pathogenic for Renal carnitine transport defect — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003060.4(SLC22A5):c.1053-6T>C, citing Invitae Variant Classification Sherloc (09022015): This sequence change falls in intron 6 of the SLC22A5 gene. It does not directly change the encoded amino acid sequence of the SLC22A5 protein. This variant is not present in population databases (gnomAD no frequency). This variant has been observed in individual(s) with primary carnitine deficiency (internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 957579). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic.
