Uncertain significance for Hypermethioninemia with deficiency of S-adenosylhomocysteine hydrolase — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000687.4(AHCY):c.685G>A (p.Ala229Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine with threonine at codon 229 of the AHCY protein (p.Ala229Thr). The alanine residue is moderately conserved and there is a small physicochemical difference between alanine and threonine. This variant is present in population databases (rs767066341, ExAC 0.002%). This variant has not been reported in the literature in individuals with AHCY-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Not Available").
