This variant lies in the OPA1 gene (transcript NM_130837.3) at 3 bases into the intron immediately after coding-DNA position 1935, where A is replaced by G. Submitter rationale: Variant summary: OPA1 c.1770+3A>G alters a conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 4e-06 in 251264 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1770+3A>G has been reported in the literature in individuals affected with OPA1-Related Disorders.
