This variant lies in the GUCY2D gene (transcript NM_000180.4) at coding-DNA position 2576, where C is replaced by T; at the protein level this means replaces proline at residue 859 with leucine — a missense variant. Submitter rationale: This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 859 of the GUCY2D protein (p.Pro859Leu). This variant is present in population databases (rs374515716, gnomAD 0.02%). This missense change has been observed in individual(s) with inherited retinal dystrophy (PMID: 26047050, 31630094). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 956847).
