This variant lies in the TTC8 gene (transcript NM_144596.4) at coding-DNA position 909, where G is replaced by C; at the protein level this means replaces glutamic acid at residue 303 with aspartic acid — a missense variant. Submitter rationale: This sequence change replaces glutamic acid with aspartic acid at codon 293 of the TTC8 protein (p.Glu293Asp). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and aspartic acid. This variant also falls at the last nucleotide of exon 10, which is part of the consensus splice site for this exon. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with TTC8-related conditions.
