Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_024426.6(WT1):c.1052C>T (p.Thr351Met), citing Ambry Variant Classification Scheme 2023: The p.T346M variant (also known as c.1037C>T), located in coding exon 6 of the WT1 gene, results from a C to T substitution at nucleotide position 1037. The threonine at codon 346 is replaced by methionine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
