Uncertain significance for Developmental and epileptic encephalopathy, 31A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004408.4(DNM1):c.2537G>C (p.Arg846Pro), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DNM1 gene (transcript NM_004408.4) at coding-DNA position 2537, where G is replaced by C; at the protein level this means replaces arginine at residue 846 with proline — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with DNM1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant is not present in population databases (ExAC no frequency). This sequence change replaces arginine with proline at codon 846 of the DNM1 protein (p.Arg846Pro). The arginine residue is weakly conserved and there is a moderate physicochemical difference between arginine and proline.
