Uncertain significance — the classification assigned by Ambry Genetics to NM_144670.6(A2ML1):c.1081A>G (p.Ile361Val), citing Ambry Variant Classification Scheme 2023: The p.I361V variant (also known as c.1081A>G) is located in coding exon 11 of the A2ML1 gene. The isoleucine at codon 361 is replaced by valine, an amino acid with highly similar properties. This change occurs in the first base pair of coding exon 11. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
