Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000038.6(APC):c.5781del (p.Gln1928fs), citing Ambry Variant Classification Scheme 2023: The c.5781delT pathogenic mutation, located in coding exon 15 of the APC gene, results from a deletion of one nucleotide at nucleotide position 5781, causing a translational frameshift with a predicted alternate stop codon (p.Q1928Rfs*42). This variant has been observed in individuals with a personal and/or family history that is consistent with APC-associated disease (Ambry internal data; Moriichi K et al. Int J Colorectal Dis, 2020 Oct;35:1967-1972; Tanabe H et al. Mol Genet Genomic Med, 2020 Sep;8:e1348). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
