NM_001429.4(EP300):c.4026-9A>G was classified as Pathogenic for Rubinstein-Taybi syndrome due to EP300 haploinsufficiency by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the EP300 gene (transcript NM_001429.4) at 9 bases into the intron immediately before coding-DNA position 4026, where A is replaced by G. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site, but this prediction has not been confirmed by published transcriptional studies. This variant has been observed in individual(s) with clinical features of Rubinstein-Taybi syndrome (PMID: 27648933, Invitae). In at least one individual this variant was found to be de novo. This variant is not present in population databases (ExAC no frequency). This sequence change falls in intron 24 of the EP300 gene. It does not directly change the encoded amino acid sequence of the EP300 protein.
