This variant lies in the ATP6AP2 gene (transcript NM_005765.3) at coding-DNA position 469, where C is replaced by T; at the protein level this means replaces arginine at residue 157 with cysteine — a missense variant. Submitter rationale: This sequence change replaces arginine with cysteine at codon 157 of the ATP6AP2 protein (p.Arg157Cys). The arginine residue is highly conserved and there is a large physicochemical difference between arginine and cysteine. This variant is present in population databases (rs138458908, ExAC 0.01%). This variant has not been reported in the literature in individuals with ATP6AP2-related conditions.
