Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014014.5(SNRNP200):c.3824G>A (p.Trp1275Ter), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with SNRNP200-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. The current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in SNRNP200 cause disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change creates a premature translational stop signal (p.Trp1275*) in the SNRNP200 gene. It is expected to result in an absent or disrupted protein product.
