NM_004281.4(BAG3):c.485C>T (p.Pro162Leu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the BAG3 gene (transcript NM_004281.4) at coding-DNA position 485, where C is replaced by T; at the protein level this means replaces proline at residue 162 with leucine — a missense variant. Submitter rationale: The p.P162L variant (also known as c.485C>T), located in coding exon 2 of the BAG3 gene, results from a C to T substitution at nucleotide position 485. The proline at codon 162 is replaced by leucine, an amino acid with similar properties. This alteration has been reported in a sudden unexplained death cohort; however, clinical details were limited (Lin Y et al. Circ Cardiovasc Genet, 2017 Dec;10:[ePub ahead of print]). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
