Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_005228.5(EGFR):c.3405C>A (p.Asn1135Lys), citing Sema4 Curation Guidelines. This variant lies in the EGFR gene (transcript NM_005228.5) at coding-DNA position 3405, where C is replaced by A; at the protein level this means replaces asparagine at residue 1135 with lysine — a missense variant. Submitter rationale: The EGFR c.3405C>A (p.N1135K) variant has not been reported in the literature to our knowledge. It was observed in 12/34564 chromosomes of the Latino subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 954844). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
