Uncertain significance for Rhabdoid tumor predisposition syndrome 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003072.5(SMARCA4):c.50C>T (p.Ser17Phe), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SMARCA4 gene (transcript NM_003072.5) at coding-DNA position 50, where C is replaced by T; at the protein level this means replaces serine at residue 17 with phenylalanine — a missense variant. Submitter rationale: This sequence change replaces serine with phenylalanine at codon 17 of the SMARCA4 protein (p.Ser17Phe). The serine residue is highly conserved and there is a large physicochemical difference between serine and phenylalanine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with SMARCA4-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
