This variant lies in the TLR3 gene (transcript NM_003265.3) at coding-DNA position 2068, where C is replaced by A; at the protein level this means replaces leucine at residue 690 with isoleucine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with TLR3-related conditions. This variant is present in population databases (rs540959112, ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
