Uncertain significance for Primary ciliary dyskinesia 32 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_031924.8(RSPH3):c.653T>C (p.Val218Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 360 of the RSPH3 protein (p.Val360Ala). This variant is present in population databases (rs765288961, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with RSPH3-related conditions. ClinVar contains an entry for this variant (Variation ID: 954713). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
