Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006269.2(RP1):c.1361T>G (p.Leu454Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with arginine at codon 454 of the RP1 protein (p.Leu454Arg). The leucine residue is weakly conserved and there is a moderate physicochemical difference between leucine and arginine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This variant has not been reported in the literature in individuals with RP1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
