This variant lies in the GCDH gene (transcript NM_000159.4) at coding-DNA position 245, where G is replaced by C; at the protein level this means replaces arginine at residue 82 with proline — a missense variant. Submitter rationale: Variant summary: GCDH c.245G>C (p.Arg82Pro) results in a non-conservative amino acid change located in the Acyl-CoA dehydrogenase/oxidase, N-terminal domain (IPR013786) of the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251404 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.245G>C has been reported in the literature in individuals affected with Glutaric Acidemia Type 1, however no all the information had been provided for analysis (example, Liu_2015). The report does not provide unequivocal conclusions about association of the variant with Glutaric Acidemia Type 1. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
