NM_006343.3(MERTK):c.1894G>A (p.Glu632Lys) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): The E632K variant in the MERTK gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The E632K variant is observed in 7/11,572 (0.06%) alleles from individuals of Latino background in the ExAC dataset (Lek et al., 2016). The E632K variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species, and in silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret E632K as a variant of uncertain significance.
