Uncertain significance — the classification assigned by Ambry Genetics to NM_021930.6(RINT1):c.1300T>C (p.Cys434Arg), citing Ambry Variant Classification Scheme 2023: The p.C434R variant (also known as c.1300T>C), located in coding exon 9 of the RINT1 gene, results from a T to C substitution at nucleotide position 1300. The cysteine at codon 434 is replaced by arginine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
