Uncertain significance for Glycogen storage disease type III — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000642.3(AGL):c.1010A>C (p.Gln337Pro), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with proline at codon 337 of the AGL protein (p.Gln337Pro). The glutamine residue is moderately conserved and there is a moderate physicochemical difference between glutamine and proline. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with AGL-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
