Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.575G>C (p.Cys192Ser), citing Ambry Variant Classification Scheme 2023: The p.C192S variant (also known as c.575G>C), located in coding exon 6 of the PMS2 gene, results from a G to C substitution at nucleotide position 575. The cysteine at codon 192 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
