Pathogenic for Capillary malformation-arteriovenous malformation syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002890.3(RASA1):c.365C>A (p.Ser122Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the RASA1 gene (transcript NM_002890.3) at coding-DNA position 365, where C is replaced by A; at the protein level this means converts the codon for serine at residue 122 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This variant is not present in population databases (ExAC no frequency). For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in RASA1 are known to be pathogenic (PMID: 24038909). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. This variant has been observed in a family affected with RASA1-related disease (PMID: 24038909). This sequence change creates a premature translational stop signal (p.Ser122*) in the RASA1 gene. It is expected to result in an absent or disrupted protein product.
