NM_000548.5(TSC2):c.900C>T (p.Gly300=) was classified as Uncertain Significance for Tuberous sclerosis syndrome by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the TSC2 gene (transcript NM_000548.5) at coding-DNA position 900, where C is replaced by T; at the protein level this means the protein sequence is unchanged (glycine at residue 300 retained) — a synonymous variant. Submitter rationale: This variant is located in the TSC2 protein. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with tuberous sclerosis complex in the literature. This variant has been identified in 1/31392 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
