Uncertain significance for Pheochromocytoma/paraganglioma syndrome 5; Mitochondrial complex II deficiency, nuclear type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004168.4(SDHA):c.131C>A (p.Ser44Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with tyrosine at codon 44 of the SDHA protein (p.Ser44Tyr). The serine residue is highly conserved and there is a large physicochemical difference between serine and tyrosine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with SDHA-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C25"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
